Mutation detailsA loxP site, a minigene consisting of the cDNA sequence for exons 11-17, a STOP cassette, a neomycin resistance gene cassette and a second loxP site were inserted into intron 10 and endogenous exon 11 was modified to change lysine codon 375 (AAG) to methionine (ATG) (p.K375M). This allele expresses the wildtype transcript and only after Cre-mediated deletion of the minigene-STOP-neo cassette will it express the mutated transcript. The mutation renders the encoded protein kinase-dead. (J:303935)
