Mutation detailsThe targeting construct contains the Myf5 open reading frame (ORF), a Kozak sequence to allow protein translation initiation. A T2A self-cleaving peptide, E2Crimson fluorescent reporter sequence, and 3 nuclear localization signals (3xNLS) inserted after exon 3, just upstream of the 3' untranslated region (UTR), of the Myf5 gene. An FRT-flanked puromycin selection cassette was also included after the 3' UTR. Flp-mediated recombination removed the puromycin cassette. (J:339277)
