Mutation detailsA loxP site was inserted into intron 2 and a STOP cassette, FRT site flanked neomycin resistance gene cassette, a second loxP site and a copy of the genomic sequence from the end of intron 2 through exon 6 (with a modified exon 5) were inserted downstream of the gene. The modification in the inserted exon 5 copy entails changing tyrosine codon 195 (TAT) to histidine (CAT) (p.Y195H). This mutation, in the ITAM motif of the encoded peptide, is the equivalent of the human p.Y196H mutation associated with diffuse large B-cell lymphoma (DLBCL). The neo cassette was removed through subsequent Flp-mediated recombination. This allele expresses the wild-type transcript/peptide and only after Cre-mediated deletion of the endogenous exons and STOP cassette will it express the modified transcript/peptide. (J:330627)
