Mutation detailsUsing the CRISPR/Cas9 method, a targeting construct consisting of an AS-loxP/NeoSTOP/loxP-murine Mecom cDNA followed by IRES-EGFP-pA fusion gene inserted into intron 1 of the ROSA26 locus into STOP-eGFP-ROSA26TV plasmid was injected into 6NK7 embryonic stem cells. Mecom and EGFP are expressed after recombination by Cre recombinase driven by endogenous Gt(ROSA)26Sor promoter. The lines containing this allele showed a low recombination rate. (J:336140)
