Mutation details: A cassette containing the following elements replaced exon 3: a lox511 site, wild-type exon 3, FRT-flanked neomycin resistance gene, loxP site, exon 3 containing a glutamine to leucine substitution at amino acid 72 (Q72L) in opposite orientation, lox511 site, and loxP site. The neomycin resistance gene was removed via flp-mediated recombination. Upon expression of cre recombinase, the wild-type exon 3 will be removed and the mutated exon harboring the Q72L missense mutation will be inserted in the forward orientation.
