Mutation detailsA 217-bp fragment containing the C-terminal tag sequences (fused in frame to the last Bhlhe41 codon) and a 1.9-kb fragment containing the mouse phosphoglycerate kinase (Pgk1) promoter linked to the neomycin-resistance gene (flanked by loxP sites) was inserted into the locus by homologous recombination in ES cells. The tag sequences contained epitopes for antibodies to Flag and V5, two cleavage sites for the TEV protease, and a biotin acceptor sequence (Biotin) for biotinylation by the E. coli biotin ligase BirA.
