Mutation details: A construct containing a CAG promoter, floxed stop cassette, tdTomato CDS, 3X Mir29-3p tough decoy, and WPRE was inserted in the gene via CRISPR-cas9 mediated recombination. The Mir29-3p tough decoy contains binding sites for the members of the Mir29 family including Mir29a, Mir29b-1, Mir29b-2, and Mir29c. Cre mediated recombination removed the floxed stop cassette.qRT-PCR analysis found a 30-60% decrease in levels in the calvaria of mice carrying the allele.
