Mutation details: The targeting vector includes a) a sequence encoding an in-frame HA epitope tag within the prodomain following amino acid 61, b) sequence encoding an in-frame myc tag within the mature domain, four amino acids downstream of the S1 cleavage site, c) three point mutations in exon 4 that introduce a serine to lysine amino acid change at the S2 cleavage site (RISR-RIKR) and a new BglII site, and d) a neomycin selectable marker flanked by loxP sites upstream of exon 4. Wheareas, wild-type BMP4 is sequentially cleaved at the S1 and then the S2 site, the S2K mutation allows for simultaneous cleavage of both sites.
