To prevent missplicing, a putative splice acceptor site downstream of the neomycin cassette was mutated (gt to ca) and an additional EcoRV-site was introduced adjacent to the second loxP site to aid in Southern blot detection. The neomycin cassette was removed via flp-mediated recombination. (J:331730)
