Mutation details: Using CRISPR-Cas9 technology in Idh1em1Nbar zygote, a floxed STOP cassette (Lox-STOP-Lox) into the mouse genomic Idh1 locus upstream of a modified exon 3 engineered to contain a nucleotide substitution in codon 132 to cause the conversion of LSL-Idh1R132H allele to the LSL-Idh1R132C. Four sequence- and (R)-2HGvalidated founder lines were selected for further experimental study. This is a representative line used for the study.
