Mutation details: A cassette containing a neomycin resistance gene, flanked by LoxP sites, was inserted downstream of exon 11. The T287A mutation was introduced by site directed mutagenesis, thereby also introducing a TspRI restriction site. The neomycin cassette was subsequently deleted of the targeted ES clones by transient transfection with a flp recombinase.
