Mutation details: The CAG-a loxP site flanked neo-Westphal stop-FLAG-mouse Foxo1* cassette followed by an FRT site-framed IRESeGFP expression cassette were inserted into ROSA26 locus via homologous recombination in the ES cells. The Foxo1 cDNA with deleted transactivation domain acting as dominant-negative Foxo1 transcripts. In presence of cre recombinase, a loxP site flanked neo-Westphal stop sequence is removed to allow expression of mutated Foxo1 gene driven by CAG promoter.
