Mutation details: Using homologous recombination in ES cells, the targeting construct was designed to generate a T-to-G mutation in exon 2, changing serine codon 34 to alanine (p.S34A). A neomycin resistance gene cassette was inserted into the intron 2. The serine 34 to alanine substitution at the dermatan sulfate (DS) attachment site of decorin produces only the decorin core protein with its N-linked oligosaccharides and is unable to produce the proteoglycan form of decorin.
