Mutation details: The following elements were inserted into the locus: the CAG promoter, a loxP site, a neomycin resistance gene cassette, a STOP cassette (four poly(A) signal sequences), a second loxP site, a mutated human yes-associated protein 1 cDNA (where all five serine codons were changed to alanine to create a constitutively active form of the encoded peptide) with 5' in-frame FLAG tag and NLS coding sequence, an IRES, and the lacZ beta galactosidase reporter gene. Only after cre-mediated removal of the neo-STOP cassette will this locus express the YAP1 and lacZ genes.
