Mutation detailsBy homologous recombination in ES cells, a loxP site was inserted into the mouse Rgs3 gene at intron1 and another loxP followed by a phospho-glycerate kinase-1-neo-bpA cassette flanked by FRT was inserted into intron 5. The resulted allele has Rgs3 gene exon 2 through exon 5 coding for PDZ domain flanked by loxP followed by a FRT flanked neo selection cassette. Cre-mediated recombination in germ cells resulted in the deletion of exon 2 through exon 5. The loss of Rgs3 mRNA was confirmed by RTPCR and the loss of full-length protein was also confirmed with western blot. (J:299414)
