Mutation details: By homologous recombination in the C57BL/6J ES cells, a SA-3xFLAG-NLS-SpCas9-NLS-polyA-loxP-HSVtk-Neo-loxP cassette was inserted into the Gt(ROSA)26Sor locus. Targeted ES cells were used for blastocyst injection to obtain chimeric mice. The loxP-HSVtk-Neo-loxP cassette was removed by mating with cre deleter mice.
