Mutation detailsThe transgene consists of a fusion protein myristoylation/palmitoylation site-attached GFP (myrGFP) and DNAs encoding C-terminal cytoplasmic domains (CT) of mouse low-density lipoprotein receptor (Ldlr; amino acid residues 813862), LDLRct inserted into the start codon located in the exon 2 of the Pvalb gene so that it is under the control of the mouse Pvalb promoter. The modified BAC transgene construct DNA was purified and linearized then injected into pronuclei of B6D2F1 mouse zygotes. Five founder lines 1-5 were generated. Line 3 with the co-expression rate of GFP and PV immunoreactivities was highest and used for the study. (J:187049)
