Mutation details: The targeting vector is designed to replace exons 3-6 (encoding the C-terminal half of the CARD and the entire CARD19 C-terminus) with a betageo-puro cassette. Absence of full-length protein expression was confirmed by western blot analysis of homozygous CD8+ T cells. Subsequent RNAseq and western blotting revealed hypomorphic expression of the adjacent Ninj1 gene.
