Mutation details: The targeting vector is designed with, from 5' to 3', a floxed-STOP cassette that contains a neomycin resistance gene, a mutant form of the human ret proto-oncogene (RET) cDNA, a viral T2A sequence, and a human histone H2B fused to a constitutively active red enhanced green fluorescence protein (tdTomato). The altered Ret sequence lacks the RET extracellular domain, which signals independently of GDNF, and is found in some patients with papillary thyroid carcinomas.
