Mutation detailsRMCE retargeted ColaA1tm22Nki, to replace the F3/frt flanked PGK-Puro casette in the 3' UTR of Cola1a by a "RMCE" plasmid encoding F3-CAG-Tol-frt, by cotransfecting a flpe-expressing plasmid to facilitate recombination into the 3' UTR of the Col1a1 locus. The transgene is a F3/FRT flanked cassette containing the CAG promoter, 8 scrambled reporter proteins, an E7 epitope and a HA tag. The 8 reporters are Kaede, Katushka, Azami Green, tagBFP, mKusabira-Orange2, eGFP, Cre recombinase and Luciferase2. All reporters are scrambled by dividing them in 3 parts, the cassette first holds the C-terminal regions of all reporters followed by all break regions and lastly all N-terminal regions. All epitopes of the 8 reporters should be expressed. (J:293819)
