Mutation details: The transgene construct was inserted using recombinase-mediated cassette exchange (RMCE). The transgene construct contains the following elements: the Gt(ROSA)26Sor promoter driving the expression of the reverse tetracycline-controlled transactivator, and the tet operon tetracycline responsive element driving the expression of turboGFP green fluorescent protein reporter. The GFP gene is fused with sequence for a miR30-based shRNA targeting Renilla reniformis luciferase. Only upon supplying mice carrying this allele with doxycycline (or tetracycline) will the GFP/shRNA hybrid gene be expressed.
