Mutation details: The 3' end of the gene was targeted with a targeting vector and sgRNAs using CRISPR/Cas9 technology. The vector that was inserted just before the stop codon contained the following elements: sequence for the F2A self-cleaving peptide, a loxP site, a neomycin resistance gene cassette, a second loxP site, a modified herpes simplex virus thymidine kinase gene (TK.007), sequence for the T2A self-cleaving peptide, and the mCherry red fluorescent protein marker gene. The neo cassette was removed through subsequent cre-mediated recombination, leaving an allele expressing a tri-cistronic gene (cyclin-dependent kinase 1 + thymidine kinase + mCherry) under the control of the host gene promoter. Cells expressing TK die when exposed to ganciclovir (GCV). This allele was targeted to ES cells that were heterozygous for the Cdk1em1Nagy allele to create homozygous mutant mice.
