Mutation detailsThe 3' end of the gene was targeted with a targeting vector and sgRNAs using CRISPR/Cas9 technology. The vector that was inserted just before the stop codon contained the following elements: sequence for the F2A self-cleaving peptide, the 5' end of a modified herpes simplex virus thymidine kinase gene (TK.007), 5' terminal repeat (TR) sequence, a neomycin resistance gene cassette, 3' TR sequence, the 3' end of the TK gene, sequence for the T2A self-cleaving peptide, and the mCherry red fluorescent protein marker gene. The neo cassette was removed through subsequent transposase-mediated excission, leaving an allele expressing a tri-cistronic gene (cyclin-dependent kinase 1 + thymidine kinase + mCherry) under the control of the host gene promoter. Cells expressing TK die when exposed to ganciclovir (GCV). This allele was used to create heterozygous mutant mice. (J:268168)
