Mutation detailsThe partial open reading frame of the S2 allele (GenBank accession AF009180) of Odocoileus hemionus hemionus or mule deer prion protein was inserted into the cosSHa.Tet cosmid expression vector, replacing the tetracycline resistance gene. This expression vector contains a 49 kb DNA fragment encompassing the Syrian hamster Prnp gene, including its promoter, with the coding region in exon 2 replaced with a tetracycline resistance gene. To increase transgene expression, the transgene nucleotide sequence was modified by site-directed mutagenesis to produce a consensus Kozak translation initiation sequence immediately upstream of the initiating ATG. Two founders, 1534 and 1536, were generated by microinjection of the transgene into fertilized embryos from Prnp knockout mice Prnptm1Cwe on an FVB/N background. (J:94219)
