Mutation details: A Y470A mutation was introduced into exon 13 of the locus, which is within the deacetylase activation domain of the protein product. A floxed neomycin selection cassette was also placed about downstream from the targeted exon. Founder mice derived from correctly targeted ES cells were crossed with EIIa-cre transgenic mice to remove the neomycin selection cassette. The introduced mutation was predicted to disrupt interactions between the Ncor2 protein product and the histone deacetylase Hdac3. Expression of the mutant allele was similar to endogenous expression as determined by immunoblot analysis of homozygous liver extracts. The mutant Ncor2 protein failed to interact with Hdac3 protein as determined by immunoprecipitation assays.
