Mutation details: A construct consisting of a floxed neo cassette, tTA site, a CMV promoter with a tetracycline responsive element, a human APOE_i4 cDNA, an IRES site and an EGFP gene, was inserted via homologous recombination. In the absence of Cre-mediated recombination the human protein is not expressed. Following Cre-mediated recombination the human protein is expressed and this expression may be suppressed with the addition of doxycycline.
