Mutation detailsThe allele with floxed minigene (exons 4-22) encoding the wild type C-terminus of Card14 has been inserted between exons 3 and 4 of the Card14 locus and an E138A mutation introduced into endogenous exon 5, was generated by Taconic. In the absence of Cre-mediated recombination, wild type Card14 is expressed from the early endogenous exons (starting from exon 3), and exons within the minigene. A 3xFLAG tag has been inserted at the end of the minigene to produce C-terminally 3x FLAG-tagged wild type CARD14. A STOP and human growth hormone poly (A) (hGHpA) cassette prevent aberrant read-through to the remaining endogenous locus. After Cre-mediated recombination, the minigene is excised, allowing transcription of Card14 from the endogenous exons and expression of CARD14[E138A]. The neo was removed via crossing with FLP deleter mice. (J:291840)
