Mutation details: A construct comprising the following elements was inserted into the host gene: a loxP site flanked STOP cassette, improved estrogen receptor nuclear translocation domain sequence fused to the cDNA for the fungal I-PpoI homing endonuclease, followed by an IRES and the EGFP fluorescent reporter gene. This creates a conditional-ready allele that will only be expressed after cre-mediated deletion of the STOP cassette and only after application of tamoxifen will the expressed endonuclease be transported to the nucleus where it targets DNA.
