Mutation detailsA construct containing a floxed minigene cassette (made of 375 bp of intron 14, mouse Braf exons 15-19, 3 polyA sites and a synthetic polyadenylation signal) a mutated version of exon 15 where the TVKS motif was changed to AVKA, and an FRT flanked neo cassette was inserted replacing the endogenous exon 15. Flp mediated recombination removed the neo cassette. Cre-mediated recombination removed the floxed minigene resulting in production of a mutant transcript. The mutations of T599 and S602 to alanines blocks phosphorylation at these residues. (J:229687)
