Mutation details: The following elements were inserted in intron 1 of the host locus: a loxP site flanked STOP cassette, the Ripk3 coding sequence with a FLAG tag fused to the 5' end and two FKBPF36V tags to the 3' end, sequence coding for the Thosea asigna virus 2A (T2A) self-cleaving peptide sequence, and the mCherry red fluorescent marker gene. This creates a conditional-ready allele that, after the removal of the stop cassette through cre-mediated recombination, would express the FLAG- and FKBP-tagged Ripk3 peptide and the mCherry reporter.
