Mutation
description
Allele Type:    Endonuclease-mediated (Null/knockout)
Mutation:    Intragenic deletion

Mutation detailsExon 3 was targeted with the CRISPR/Cas9 system. The sgRNA sequence was 5'-CACCTAGCCTCGCACGAActt-3' (recognition sequence in upper case) and the experiment resulted in a 5 bp deletion (CTCGC) in the middle of the recognition sequence. The resulting frameshift introduces a premature stop codon soon after, knocking the gene out. Western blots confirmed the absence of peptides translated from this allele in homozygous mice.
