Mapping and Phenotype information for this QTL, its variants and associated markersJ:221322The protozoan Trypanosoma cruzi, the causative agent of Chagas disease in humans, parasitizes several other mammalian species. After infection with T. cruzi, the parasites survive and multiply in nucleated cells as amastigotes, eventually reaching the bloodstream as trypomastigote forms. The acute infection phase is characterized by high levels of circulating parasites, while parasite proliferation is contained during the chronic phase. In this study the authors analyzed the relationships between the genetic controls involved in antibody production and inflammatory responses and resistance to T. cruzi infection by examining the course of parasite infection in mice selected for high (HIII) and low (LIII) antibody responses or for maximal (AIRmax, referred to as Ibut:AIRH) and minimal (AIRmin, referred to as Ibut:AIRL) acute inflammatory responses (AIR). F1 (HIII LIII) hybrids, obtained by reciprocal crosses (HIII males x LIII females or HIII females x LIII males), showed identical resistance patterns to T. cruzi challenges, indicating that there were no maternal or parental effects in determining resistance/susceptibility to infection. However, there was an overdominance effect of the resistance phenotype, with F1 hybrids showing greater resistance than the parental HIII line. A segregating population of 242 mice was constructed by backcrossing (HIII x LIII) F1 mice with the susceptible parental LIII line to create an F2 segregating backcross [F1(HIII LIII) x LIII]. Composite data related to the survival times of males and females from this backcross population were submitted to genetic analysis, considering sex as a covariate for the trait. Two QTL relating to Trypanosoma cruzi susceptibility were identified on Chromosome 1 at D1Mit303 (62,907,596 - 62,907,723 bp; 34.8 cM):The first locus, Tcsm (Trypanosoma cruzi susceptibility, mortality), has a LOD score of 1.6 (LRS score = 7.4).The second locus, Tcsst (Trypanosoma cruzi susceptibility, survival time), has a LOD score of 1.97 (LRS score = 9.1).No specific candidate gene or allele information was provided by the authors. |
