Mutation detailsA beta galactosidase (lacZ) and a floxed luciferase reporter gene cassette were inserted between the penultimate codon and the stop codon in exon 3 and an FRT site flanked puromycin resistance gene cassette was inserted downstream of exon 3. The reporter gene sequences were preceded by sequence coding for self-cleaving T2A peptide. The puro cassette was removed through flp-mediated recombination, yielding a dual reporter allele. Western blots showed expression of the gene from this allele to be considerably lower than from the wild-type allele. (J:237855)
