Mutation detailsThis transgene comprises the cytomegalovirus (CMV) promoter, a 6564-base pair mouse Cux1 cDNA containing 27 bp of 5' untranslated region (5'-UTR), the 4544 bp protein-coding sequence and 1993 bp of 3'-UTR, and the SV40 small t intron and polyadenylation signal. CUX1 protein is detected at low levels in nuclear extracts of kidney and spleen of adult wild-type mice and at high levels in kidney, liver, spleen and testis of transgenic mice by immunoblot analysis using a goat polyclonal antibody to a 19-amino acid N-terminal peptide of CUX1, which is not present in the truncated testis-specific endogenous CUX1 protein isoform. Immunohistologic analysis reveals ectopic expression in capillary loop-stage and maturing glomeruli of transgenic embryos and in both glomeruli and tubular cells of adult transgenic mice. (J:76261)

Notes
Three transgenic founder mice transmitted the transgene to their offspring. The two lines with the highest kidney expression, designated 2189 and 2200, were subjected to further analysis and reported to have identical phenotypes. The third line, 2235, exhibited no differences vs. wild-type littermates and was not studied further. (J:76261)
