• Mutation details: The following elements comprise the conditional by inversion (COIN) sequences that were inserted: lox2372 and loxP sites in intron 1, and, in reverse orientation, FRT site, a neomycin resistance gene cassette, FRT site, loxP site, lox2372 site, rabbit HBB2 intron 2 splice acceptor site, sequence coding for T2A self-cleaving peptide, far-red fluorescent protein FP635 cDNA, and rabbit HBB2 3' UTR (for poly(A) signal) in intron 3. Flp-mediated recombination removed the neo cassette. Cre-mediated recombination would invert the inserted sequence, which includes exons 2 and 3, via the loxP sites and delete the first loxP site via lox2372, creating an irreversible reporter knockout allele. The allele was created in ES cells containing a Gt(ROSA)26Sor allele. (J:217084)
