Mutation details: A lacZ gene with a nuclear localization signal (nls-lacZ) was placed under the control of the TP1 promoter, which consists of 12 multimerized RBP-Jkappa binding motifs upstream from a minimal beta-globin promoter. The TP1 promoter is transactivated in a RBP-Jkappa-dependent manner by the activated forms of the four mammalian Notch receptors, and can be used to detect Notch pathway activity. Only one transgenic line generated was shown to express lacZ and was expanded and studied further.
