Mutation detailsThe targeting construct inserted into the locus a CAG promoter, floxed membrane-targeted tandem-dimer Tomato (mT) reporter gene, an improved sensor cGMP indicator (cGi500; containing an FRET-activated, cGMP-sensitive ECFP/EYFP fusion) and an FRT-flanked neomycin resistance cassette. Cre-mediated recombination can remove the Tomato reporter gene to allow the expression of the cGMP indicator in a time- and cell type-specific manner. The cGMP indicator has an EC50 of 500 nmol/L.
