Mutation detailsThe transgene contains a 516-bp promoter fragment of the stress-inducible mouse Hspa1b gene (also called Hsp70), including 284 bp of 5' flanking sequence and the 5' untranslated region, separated by a Kozak consensus sequence (ACCACC) and a rabbit beta-globin intron from the mPlum open reading frame, which is followed by the SV40 early polyadenylation signal. Brains of transgenic mice in which focal ischemia had been induced one day prior exhibited a fluorescence resonance imaging (FRI) signal coincident with expression of endogenous Hspa1b as determined by immunohistochemistry, and mPlum fluorescence was observed by in vivo confocal microscopy in penumbra neurons and in blood vessels at the ischemic core. mPlum expression does not affect neuronal or penumbral viability.
