Mutation detailsThis transgene employs the Fabpl4x at -132 promoter cassette from the pTS4 expression vector to drive expression of a full-length cDNA encoding chicken netrin 1 (NTN1) with a C-terminal Myc epitope tag (EQKLISSEEDL). The promoter cassette contains nucleotides (nt) -596 through +21 of the rat fatty acid binding protein 1, liver (Fabp1) gene with an insertion at nt -132 -- just upstream of its endogenous position -- of four additional tandem copies of the 35-bp putative nuclear protein binding site, composed of a direct repeat of the sequence TTCTGNNTT separated by five nts, that resides at nts -172 to -133 of the Fabpl gene. Notes
Five lines of transgenic mice, designated Tg-netrin-1/1 through 5, were analyzed. Data for individual lines is distinguished only on the Southern blot demonstrating presence of the transgene in each line. Otherwise, numerical data are pooled, and line numbers are not associated with either figures or text descriptions. Mice bearing this construct were shown to express transgene-derived transcripts and/or protein throughout the gut epithelium in both colon and small intestine - in both villi and crypts dissected from the latter - and in the kidney, but not in the lung.
