Mutation detailsThis transgene contains the regulatory sequences of the mouse vav 1 oncogene (Vav), which is expressed ubiquitously and exclusively in hematopoietic cells, driving expression of the human tumor-associated NPM1/ALK fusion protein. This chimeric tyrosine kinase, comprising the N-terminal 40% of the ubiquitously expressed protein nucleophosmin (nucleolar phosphoprotein B23, numatrin) (NPM1) joined to the entire cytoplasmic tail of anaplastic lymphoma kinase (ALK), is expressed in tumors from a fusion gene resulting from the t(2;5)(p23;q35) chromosomal translocation that is present in 40-50% of anaplastic large cell lymphomas. Line 1 bears a single copy of the transgene, as estimated by Southern blot analysis. RT-PCR analysis of mRNA and immunoblot analysis of tissue lysates demonstrated the presence of transgene-derived transcripts and protein, respectively, in tumors, but not in normal thymus or spleen, of transgenic mice.
