Mutation detailsIn this transgene construct, the Wnt1 promoter drives lacZ cDNA, which is followed by the Wnt1 enhancer. Between the promoter and lacZ sequence is a loxP-flanked PGKneo cassette. This stop cassette prevents expression of lacZ in the absence of cre-mediated recombination. Two lines were obtained. R170 has 2 copies of the transgene, while R44 has 3 copies of the reporter. Both lines displayed the expected Wnt1 pattern after E11.5 when crossed to reporter animals. The R170 line showed stronger recombination activity and was used for further studies. (J:69326)
