Mutation details: Mice bearing this transgene express luciferase from a promoter that is activated by glioma-associated oncogene proteins (GLI), primary targets of sonic hedgehog signaling. The transgene is derived from the pGL3b/8GliBS-luc plasmid, which contains eight copies of the human GLI-binding DNA consensus sequence (5'-GAACACCCA-3') upstream of the chicken lens delta-crystallin (ASL/CRYD) basal promoter driving expression of the firefly luciferase gene. The transgene serves as a reporter of GLI activity based on bioluminescence (BLI) following local or systemic luciferin injection. Nine founders with germline transmission were found; granule cell preparations from only one line displayed tight regulation of expression. Transgenic line numbers are not provided.
