Mutation details: The tetracycline transactivator (tTA) coding sequence is driven by the 3.6-kb promoter from the rat collagen, type 1, alpha 1 gene, which drives expression in both mature and immature osteoblasts. The coding sequence is followed by the rabbit beta-globin intron and a polyadenylation signal. In the absence of tetracycline, expression of a second transgene whose promoter is regulated by the tet operator sequence (tetO) is induced; this induction is suppressed by tetracycline administration.
