Mutation details: The transgene comprises the upstream promoter/enhancer region from the mouse interleukin 2 gene extending from -8.4 kb through nucleotide +45 followed by a cDNA encoding enhanced green fluorescent protein (EGFP) and the 3' splice and polyadenylation signals from a human beta-globin gene. EGFP expression is inducible, efficient and specific to the tissue/cell types and developmental stages where IL2 normally is expressed, i.e., in peripheral T cells and in immature thymocytes and thymocytes undergoing positive selection. It was estimated by Southern blot hybridization that thirty-three copies of the transgene construct had integrated into the genome of this line.
