Mapping and Phenotype information for this QTL, its variants and associated markers


Chromosome substitution strains derived from A/J donor chromosomes on a C57BL/6J genetic background were used to map QTL associated with 90 phenotypes including blood, bone and metabolic traits. Intercrosses between (C57BL/6J-Chr 6A/J/NaJ x C57BL/6J)F2 and (C57BL/6J-Chr 10A/J/NaJ x C57BL/6J)F2 mapped several obesity and liver triglyceride QTL on chromosomes 6 and 10. Animals were placed on a high fat diet at 5 weeks of age and weighed every 2 weeks for 100 days. Background strain C57BL/6J displays increased body weight and liver triglyceride levels compared to donor strain A/J.

Two traits were explored in congenic strains containing nested series of segments derived from chromsomes 6 and 10; CSS-6 and -10 for diet-induced obesity and CSS-10 for levels of liver triglycerides.

Obrq1 (obesity resistance QTL 1), Obrq2, Obrq3 and Obrq4 mapped to mouse Chromosome 6. The order of the QTL is centromere - Obrq2 - Obrq3 - Obrq4 - Obrq1 - telomere.

Obrq2 was mapped in 6C2 (B6.A-(D6Mit138-D6Mit159)/6C2Na) and was linked with D6Mit159 within the congenic interval.

Obrq3 was mapped in 6C3 (B6.A-(D6Mit138-D6Mit223)/6C3Na) and was linked with D6Mit223 within the congenic interval.

Obrq4 was mapped in 6C4 (B6.A-(D6Mit138-D6Mit274)/6C4/Na and was linked D6Mit274 within the congenic interval.

Obrq1 was mapped in 6C12 (B6.A-(D6Mit284-D6Mit15)/6C12Na) and was flanked within the congenic interval by D6Mit284 and D6Mit287 within the interval. [Fig.3, SI]

QTL identified on the Chromosome 10:

Obrq9 (obesity resistance QTL 9) was mapped in 10C1 (B6.A-rs29331213/10C1Na) linked with rs29331213 within the interval.

Obrq10 was mapped in 10C2 (B6.A-(rs13480792-rs29331213)/10C2Na) and is linked with rs13480792 within the congenic interval.

Obrq11 was mapped in 10C6 (B6.A-(D10Mit230-rs29331213)/10C6Na) flanked by markers D10Mit230 and D10Mit95 within the congenic interval.

Obrq12 was mapped in 10C7 (B6.A-(rs13480651-rs29331213)/10C7Na) flanked by markers rs13480651 and D10Mit42 within the congenic interval.

Ltgq1 was mapped in 10C9 (B6.A-(rs29359333-rs29375724)/10C9Na) linked with rs29359333.

Ltgq2 (liver triglyceride QTL 1) was mapped in 10C1 (B6.A-rs29331213/10C1Na)and was also linked with rs29331213.

The order of liver triglyceride and and obesity QTL is centromere - Ltgq1 -Obrq12 - Obrq11 - Obrq10 - Obrq9/Ltgq2 - telomere. [ Fig S2 and SI].
