Mapping and Phenotype information for this QTL, its variants and associated markers

Quantitative trait loci (QTL) for dorsal and ventral hippocampal volume in mice were previously mapped in BXD and AXB recombinant inbred (RI) strains (J:117408). Suggestive linkage to ventral hippocampal volume was identified at 33 cM on mouse Chromosome8 near D8Mit31 (LOD=2.8) and 31 cM on mouse Chromosome 13 near D13Mit94 (LOD=2.7). These loci were examined in the current study for association between ventral hippocampal volume and behavioral tasks such as working memory and anxiety. The chromosome 8locus is designated Vnhv1 (ventral hippocampal volume 1) and the chromosome 13 locus is designated Vnhv2 (ventral hippocampal volume 2). SNP marker 13.067.300 was used to genotype Vnhv2 in this study.

Male animals from ten BXD RI strains were tested at 4-5 months of age. The five BXD RI strains displaying small ventral hippocampal volumes carried DBA/2J-derived alleles at Vnhv1 and C57BL/6J-derived alleles at Vnhv2 (genotype A). Conversely, the five BXD RI strains displaying large ventral hippocampal volumes carried C57BL/6J-derived alleles at Vnhv1 and DBA/2J-derived alleles at Vnhv2 (genotype B).

An association between working memory and genotype was observed. RI strains with large ventral hippocampus volumes (genotype B) had a significantly larger number of working memory errors during trial 3 of the water mazetest compared to RI strains with small ventral hippocampus volumes (genotype A).

Anxiety was also associated with RI strain genotype. The small hippocampal volume RI strains (genotype A) displayed decreased locomotor activity in the center of an open field compared to large hippocampal volume RI strains (genotype B). However, the total number of entries into the center of an open field was not significantly correlated to genotype at Vnhv1 or Vnhv2.

Referencememory, acoustic startle response, prepulse inhibition and elevated plus maze performance did not display association to RI strain genotype.
