Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 457 (EMH x EML)F2 animals to identify QTLs associated with adiposity traits. Genome scan was performed using 87 polymorphic markers at a resolution of 23.6 cM. F2 animals were phenotyped for brown and white adipose depots at 18 weeks of age. Parental strains EMH and EML are high- and low-food intake selection lines, respectively. Highly significant QTLs (P<0.001) are reported below.

Mfat1 (M-line adipose tissue, general adiposity 1) maps to mouse Chromosome 4 between 47cM - 53 cM. This locus is linked to all brown and white adipose tissue measures. Lepr (46.7 cM) is a potential candidate gene for Mfat1. Previously identified obesity QTLs near Mfat1 include Pfat1 (30 cM), Adip11 (46 cM), Dob1 (50 cM), Afpq2 (66 cM), Abfw2 (64 cM), and Tafat (70 cM).

Mfat2 (M-line adipose tissue, white adipose tissue 2) maps to mouse Chromosome 9 between 17 cM - 50 cM. This locus is female-specific and shows linkage to white adipose tissues measures (retroperitoneal, gonadal, mesenteric, subcutaneous). Mfat2 explains 8.6% of the gonadal fat pad variance, 7.4% of the retroperitoneal fat pad variance, and 5.1% of the subcutaneous fat pad variance. Apoc3 at 27 cM is a potential candidate gene for Mfat2. Previously identified obesity QTLs mapping near Mfat2 include Obq5 (19 cM), Afw4 (29 cM), Mob8 (42 cM), Adip5 (42 cM), Dob2 (60 cM), Adip14 (60 cM), and Obq18 (65 cM).

Mfat3 (M-line adipose tissue, abdominal and gonadal fat 3) maps to mouse Chromosome 11 between 1 cM - 9 cM. This locus islinked to retroperitoneal and gonadal white adipose tissue measures. Gcgr was named as a potential candidate gene for Mfat3. Afw5 (10 cM) is a previously identified obesity QTL located near Mfat3.

Mfat4 (M-line adipose tissue, abdominal and gonadal fat4)maps to 0 cM on mouse Chromosome 12. This locus shows linkage to retroperitoneal and gonadal white adipose tissue measures.

Mfat5 (M-line adipose tissue, general adiposity 5) maps to 65 cM on mouse Chromosome 13. The Mfat5 QTL interval spans 46 cM to the terminal end. This locus is linked to brown adipose tissue measures, gonadal and subcutaneous white adipose tissues measures, and total fat depot measures. Nnt (64 cM) and Cart are potential candidate genes for Mfat5.

Mfat6 (M-line adipose tissue, general adiposity 6) maps to 66 cM on mouse Chromosome 15 and is linked to total fat depot measures. Mfat6 shows suggestive linkage to subcutaneous white adipose tissue.

Mfat7 (M-line adiposity tissue, white adipose tissue 7) maps to mouse Chromosome 19. This locus is linked to white adipose tissue measures. Mfat7 explains 4.8% of the retroperitoneal fat depot variance, 4.2% of the mesenteric fat depot variance, and 5.5% of the subcutaneous fat depot variance. The Mfat7 QTL interval spans 15 cM - 62 cM.Previously identified obesity QTLs mapping near Mfat7 include Abfw4 (43 cM), Nobq2 (47 cM), and Obqw5 (47 cM). No feasible candidate genes have yet been identified for Mfat7.

Mfat8 (M-line adiposity tissue, general adiposity 8) maps to mouse Chromosome X between 9 cM and 53 cM. Linkage to retroperitoneal white adipose tissues peaks at 38 cM. Linkage to mesenteric white adipose tissue peaks at 17 cM, and linkage to total fat mass peaks at 40 cM. A previously identified obesity QTL named Fob4 (37 cM) overlapswith Mfat8 and may represent the same locus. Other overlapping obesity QTLs include Obq6 (16 cM), Aftw11 (17 cM), Obq22 (34 cM), Obwq5 (52 cM), and Dob7 (59 cM). Brs3 was named as a potential candidate gene for Mfat8.
