Mutation detailsA loxP flanked stop cassette was inserted downstream of the proximal promoter into the 5'UTR. To stop transcripts from the distal promoter, the cassette included a 1.7 kb splice acceptor (SA) sequence from the mouse Engrailed-2 gene. The NLS was placed in-frame between the upstream distal promoter exons and the Betagal gene, and transcription stop elements were included downstream of lacZ to prevent read-through of the cassette. The neo was excised by transient expression of cre recombinase. (The Engrailed2-SA-lacZ inserted sequence is flanked by loxP sites.) (J:106769)
