Mutation details: The transgene comprises a cDNA encoding the enzymatically nonfunctional S145G mutant human hepatic lipase (uniprot E7EUK6; NM_000236.3:c.616A>G;p.Ser206Gly) cloned between the promoter and polyadenylation signal of the human apolipoprotein E (APOE) gene; it also includes the hepatic and macrophage control regions from the Apoe/Apoc1 locus. Northern blot analysis demonstrated expression of the transgenic mRNA in liver, but not in any of 8 other organs/tissues examined. Western blot analysis revealed presence of the human protein in post-heparin plasma. The level of both mRNA and protein expression is 3- to 5-fold that of endogenous gene expression in wild-type mice.
