A genome wide search was made in an attempt to find microsatellite markers linked to Ccs1 (colon cancer susceptibility 1) in 122 progeny derived from a (ICR/Ha x C57BL/6Ha)F1 x C57BL/6Ha backcross. ICR/Ha is susceptible and C57BL/6Ha resistant to DMH induced colon tumors. 177 polymorphic markers spaced 5 cM - 30 cM apart were used for the genome scan. Chi square tests indicated linkage with all tested Chromosome 12 markers and the association of Ccs1 with D12Mit6 (45 cM) showed the greatest significance (P 0.001) A LOD score of 2.9 between Ccs1 and D12Mit6 was also indicated. ICR/Ha-derived alleles at Ccs1 are associated with colon tumor susceptibility whereas C57BL/6Ha-derived alleles are associated with colon tumor resistance. A potential candidate gene mappingnear Ccs1 is Tgfb3 (41 cM).

A linkage map of Chromosome 12 based on the backcross analysis indicated the following order: D12Nds11 - 14.5 cM - D12Mit2 - 12.9 cM - D12Mit4 - D12Mit5 - 3.2 cM - D12Mit51 - D12Rfj1 -3.2 cM - D12Mit6 - Ccs1 - 3.2 cM - D12Mit30 - 4.8 - cM - D12Mit7 - 9.7 cM - D12Mit8.
